Özet notu Piperazine derivatives, with their wide range of biological activities, are among the most significant pharmacophores in medicinal chemistry. By strategically altering the structure and placing substitutuents at different positions of the piperazine core structure, these biological functions can be controlled. As a result, piperazine derivatives have been attractive building blocks for chemical synthesis and drug design.<br/>In this study, a novel piperazine derivative was synthesized via the Mannich process under reflux reaction, wherein the N-substituted phenylpiperazine ring was reacted with 4-fluorophenyl-piperazine, to give [4-(4-Fluoro-phenyl)-piperazin-1-yl] -(4-phenyl-piperazin-1-yl)-methane. The structure mainy consists of two piperazine rings bridged by a methylene group with fluorine atom as a substituent.<br/>Proton Nuclear Magnetic Resonance Spectroscopy (1H-NMR) and Fourier Transform Infra-Red (FT-IR) validated that compound was successful synthesized. In the FT-IR spectrum, the absence of an N-H stretches suggested that the reaction occurred at position one on N-atom. Melting point analysis and Thin Layer Chromatography (TLC) were used to assess the compound’s purity. In order to understand the pysiochemical and ADME (absorption, distribution, metabolism, and excretion) properties of the synthesized molecule for drug likeness studies computationally prediction was performed by using SwissADME online software software. Additionally, in terms of antimicrobial susceptibility, the disk diffusion test showed high to moderate efficiency against gram-positive (Bacillus) and gram-negative (E. coli) organisims after 24h incubation time. These findings point that this novel compound could have potential to be further studied as a drug candidate.
